Stage-Dependent Association of Nuclear and Chloroplast DNA Molecules through Bridges Transiently Formed between the Two Organelles in Synchronized Cells of Euglena gracilis

Using a monoclonal antibody to DNA from mouse together with gold-conjugated anti-mouse IgG antibody (Au-Anti IgG) it has been possible to visualize the DNA of the nucleus, chloroplast and mitochondrion of Euglena gracilis Klebs strain Z Pringsheim in the electron microscope. At stages in light/dark synchronous cultures of Euglena at which the chloroplasts show a close temporary association with each other and with the nucleus via numerous bridges, immunoelectron microscopy confirms that interactions take place among the DNAs of different chloroplasts of the aggregate. Immunoelectron microscopy also showed that through the bridges formed between the nucleus and the chloroplasts, DNA molecules from the nuclear chromosomes meet with those from the chloroplast nucleoids. These findings indicate that genomic interactions may occur between chloroplasts and between the chloroplasts and the nucleus during the cell cycle.
